I'm using a Razer DeathAdder mouse and a BlackWidow Ultimate Stealth 2014 and have no issues.
can you attach a copy of a hijack this log so we can see what's running? Any overlay's like discord, overwolf? Something that maybe hooking the input?

just a thought
First, I tried unplugging the controller and running the game. Keyboard and Mouse both worked normally, but I couldn't plug the controller back in after the game was running and have it work.
Then I uninstalled Windows Update KB3140743. I was able to start the game with everything plugged in and everything worked normally.
Going to reinstall synapse again just to see if it plays nice.
EDIT: No problems after reinstalling synapse.

Seems that solves the problem in other games as well from what I've been reading around the net. So it appears a forced Windows 10 update broke basic input functionality. I'll mark this fixed for now and hopefully, Microsoft will fix what they broke in another update soon.

It'd be better to notify, even if it were security related.

I think there's some youtube tutorials on disabling these wonderful with no user intervention....Win 10 features.
Spybot-Anitbeacon will go through your registry every reboot and force certain privacy things off, even after updating....I have no idea if it will disable auto-updates or not though.

It was one of the first things I changed from the default setup. If I remember, it only installs the updates if you use the restart button from in the update window. The only difference I've noticed from the Windows 7 update scheme the way I have it set up, is it no longer tells me what is in each set of updates, which is kind of annoying. But other than that it appears to function the same way.

The default I think was to automatically install the updates at like 3am. But even then, since I shut my computer down each night, they would never install!

Security Update for Windows 10 KB3140768 was released a little after the update that caused this issue (I got it on 3/10). KB3140768 causes the same issue as KB3140743 with the xbox one controller preventing the keyboard and mouse from working with some games (this one included).
This issue started cropping up again, and I was looking in my installed updates to make sure they didn't stealth install the broken update again. They didn't, but this new update was there and after a quick internet search appears to be causing the same issues... I'll uninstall in and try just to confirm.
EDIT: Yep, KB3140768 causes the same issue as KB3140743!
